Description: Granzyme C is a serine protease that is found in the granules of cytotoxic T lymphocytes and natural killer cells (NKs). It induces the apoptosis of target cells by the externalization of phosphatidylserine, nuclear condensation and collapse, as well as the introduction of single-stranded DNA nicks. It can also induce apoptosis via mitochondrial swelling, which results in the loss of membrane potential. Reported studies show that granzyme C can support CTL-mediated killing in the absence of granzyme A or B. Additionally, wild-type mice have delayed upregulation of granzyme C relative to granzyme B and a mutation in the granzyme B gene shows earlier upregulation of granzyme C. Further studies show that in the event that granzyme B fails, the mouse models infected with persistent viruses have delayed expression of protection provided by granzyme C.   collapse

Description: CD158e1, also known as NKB1, is a 70 kD member of the immunoglobulin superfamily that is expressed on a subset of natural killer cells and T cells at varying levels among individuals. NKB1 is a type I membrane protein containing two immunoglobulin C2-type domains. The interaction of NKB1 with specific HLA-B antigens on a target cell (the HLA-Bw4 allele, for example) inhibits cytotoxicity and prevents target cell lysis and death. The interactions between KIR and MHC class I are thought to be important in NK and T cell regulation following antigen stimulation. The absence of ligands for KIRs may lower the threshold for activation through activating receptors and increase inflammation and susceptibility to autoimmune disease.   collapse

Description: Alpha-synuclein is expressed principally in the nervous system, but it is also produced in other tissues, including the skin. In the brain, the protein is primarily neuronal, but it is also present in glia. Neuronal α-synuclein is concentrated in presynaptic nerve terminals, interacts with plasma membrane phospholipids, and is also present in nuclei and mitochondria. At least three isoforms of α-synuclein are produced through alternative splicing. The most common isoform is a 140 amino acid-long transcript. Other isoforms include, α-synuclein-126, lacking residues 41-54; and α-synuclein-112, which lacks residues 103-130. α-synuclein’s physiological role is poorly understood, but the protein has been implicated in regulating dopamine release and transport, synaptic vesicle clustering, and functioning as a SNARE-complex chaperone. α-synuclein fibrils are a major component of the intracellular Lewy bodies that are associated with Parkinson's disease, Lewy body dementia, and multiple system atrophy. Phosphorylation of α-synuclein at Tyr39 may modulate clearance of the protein and contribute to Parkinson's disease.   collapse
