Annexin V-FITC conjugated protein binds to cell surfaces expressing phosphatidylserine, an early apotosis marker. Cells stained with propidium iodide (PI), a non-cell-permeable DNA dye, indicate necrotic cells. Cells stained with both PI and annexin V-FITC demonstrate later stage apoptosis and early necrosis. Description: The BrdU Cell Proliferation Assay Kit detects 5-bromo-2’-deoxyuridine (BrdU) incorporated into cellular DNA during cell proliferation using an anti-BrdU antibody. When cells are cultured with labeling medium that contains BrdU, this pyrimidine analog is incorporated in place of thymidine into the newly synthesized DNA of proliferating cells. After removing labeling medium, cells are fixed and the DNA is denatured with our fixing/denaturing solution. Denaturing of DNA is necessary to improve the accessibility of the incorporated BrdU to the detection antibody. A BrdU mouse mAb is then added to detect the incorporated BrdU. Anti-mouse IgG, HRP-linked Antibody is used to recognize the bound detection antibody. Chemiluminescent reagent is added for signal development. The magnitude of light emission, measured in relative light units (RLU), is proportional to the quantity of BrdU incorporated into cells, which is a direct indication of cell proliferation.   collapse

Description: The Cell Health Assay Kit is a fluorescence-based assay that determines cell viability by measuring intracellular esterase activity and plasma membrane integrity. The kit contains Calcein-AM, which stains viable cells, and Propidium Iodide (PI), an indicator of dead cells. Calcein-AM is the acetomethoxy form of calcein, a highly lipophilic, cell membrane permeable dye. Intracellular esterase converts the non-fluorescent Calcein-AM to the highly fluorescent calcein, which is retained within live cells and produces an intense green fluorescence. The DNA-binding agent Propidium Iodide is cell membrane impermeable and only enters dead cells or those with damaged cell membranes. Intracellular PI binds DNA and undergoes an approximate 40-fold enhancement in fluorescence intensity. As a result, live cells will produce a strong green fluorescence resulting from the conversion of Calcein-AM to calcein, while dead cells produce a strong red fluorescence due to the presence of Propidium Iodide. At the recommended reagent concentrations and volumes, one kit will provide for 1000 assays (96-well plate format) or 200 flow cytometry assays.   collapse
